Description
VH298
VH298 (CAS No.: 2097381-85-4, CAT No.: SRCV-003) is a highly potent, cell-permeable, and selective small-molecule inhibitor of the E3 ubiquitin ligase Von Hippel-Lindau (VHL). VH298 disrupts the interaction between VHL and hydroxylated HIF-α (hypoxia-inducible factor alpha), thereby stabilizing HIF-α and triggering a hypoxia-mimetic cellular response in normoxic conditions. This chemical probe is widely used in chemical biology and medicinal research to study the hypoxic signaling cascade and explore new pathways for therapeutic intervention.
VH298 exhibits nanomolar affinity for VHL (Kd = 80–90 nM), resulting in time- and concentration-dependent accumulation of hydroxylated HIF-α and upregulation of HIF target gene transcription at mRNA and protein levels. Studies indicate its application in wound healing, angiogenesis, and the modulation of cell proliferation, making it an attractive starting point for developing new hypoxia-activated therapeutics and investigative probes for cellular hypoxic responses.
