HATU

HATU (O-(7-Azabenzotriazol-1-yl)-N,N,N’,N’-tetramethyluronium hexafluorophosphate) is a premier coupling reagent for solid-phase and solution-phase peptide synthesis. It is especially renowned for rapid amide bond formation, high yields with difficult sequences, and minimal racemization. HATU readily reacts with carboxylic acids in the presence of an organic base (such as DIPEA) to generate activated OAt esters, accelerating acylation of both amines and alcohols. Its high reactivity is a result of the neighbouring group effect from the azabenzotriazole moiety, which facilitates the critical transition state for amide formation. HATU is soluble in DMF and other polar aprotic solvents, and is routinely used in Boc and Fmoc peptide synthesis protocols as well as in cyclization and fragment condensation chemistry.
